  • How to prepare lipid mix?

    Simple than you think! You need to only care about the quantity of each lipid, and mix! For example, if you are using Pfizer-BioNtech mix, you can simply follow the table below:

    So in total two steps you will need to take:

    1. Prepare stock solution for all lipid by disovle the lipids with pure ethanol(purity>99%). Cautious, ALC0315 is oil form, and very disovable in Ethanol, so use tips to weight.
    2. Mix all the stock lipid and add more ethanol following the table above.

    Beare in mind, we have the highest purity of all lipid: ALC0315, ALC0159, Distearoylphosphatidylcholine, and cholesterol. After all, you can store your lipid in -80C and take it whenever you want. I know you may wondering how the lipid propotion was decided. We prepared a post for you to understand the details here. You will understand how the mRNA and lipid proportion were caculated based on the literature. 

  • How to formulate the mRNA-LNP?

    Here is the video guide for formulate your mRNA-LNP with our pump. Moreover, when you are using Pfizer-BioNtech lipid mix, the best Flow rate is: mRNA:lipid mix = 8:1. In return, the partical size will be around 100nm. Regarding the best size for mRNA-LNP for delivery, we have listed the possible ranges based on the literature. But, we do think around 100nm is the best target you should aim to for your vaccine.

  • Should I remove the Ethanol from mRNA-LNP?

    Yes, you must remove the ethanol. As the LNP size is far bigger than a protein (100nm against 1-3nm), therefore you can use all the membrane based method to purify the mRNA-LNP, such as Amicon devices, we very often use Ultra-15 for 200ug mRNA. You can caculate based on your mRNA load.
